Hello Cliff!

Thanks for providing not only the problem, but plenty of documentation!
Unlike its DoubleSpace predecessors, DriveSpace 3 is inherently trouble-free. However, for it to compress a drive properly, it needs unrestricted access to the files on that drive.
The list of running programs tells me that you have good taste in software, but that there are several potential sources of conflict for DriveSpace 3.
If I had to restrict my long-distance diagnosis to one likely conflict, it would be with the McAfee anti-virus program (which I think is super, BTW). But it's SOP to disable McAfee, or any anti-virus software, before installing new software or reconfiguring a drive.
My recommendation is this:
Temporarily remove from the Startup group ALL PROGRAMS.
In your WIN.INI file, temporarily disable the RUN= and LOAD= lines by putting a semicolon in front of them. This will prevent Windows from starting programs which begin automatically but don't use the Startup group.
Now run DRVSPACE with nothing else open.
That should do it. After the drive has been compressed, you can remove the semicolons in the WIN.INI file and restore the Startup group.
